It's all you need to know if you have a Kindle. Once you have it installed on your computer (mac or pc), you can use it to send books you have acquired from places like manybooks.net exactly where they need to go on your Kindle. And it only takes a couple of clicks with your mouse.
Once you have your Kindle connected to your computer, if you already have Calibre up and running, you will see the tool bar change. It knows when there is a Kindle connected and there are a couple of extra buttons to help you with various tasks.
If you have your Kindle connected, you can quickly switch between the Library and the Device button to see a list of books. The Library shows what is on your computer. If anything listed is also on your connected Kindle, you will see a check mark next to it.
Click the Device button to see the books on your connected Kindle listed. Any books with a check mark next to them are also on your computer hard drive in the Library. I discovered (quite by accident) that if you right click any of the books listed on your Kindle, you can choose to have a copy of the book sent to your Library on your computer. How about that for an interesting, no fuss way to back up the books on your Kindle?

Kindle for Devices... New and Improved!
Collections!
If you have fallen in love with your collections on your Kindle, you will appreciate finally having them in your Kindle for your PC or Mac. When you install the latest update, you will see a choice to import your collections from your Kindle (or Kindles if you have more than one). Now, the actual books won't be imported, but placeholder/shortcuts to the titles in each collection you have will be listed. Clicking them will cause each to be downloaded as you want them.
I LOVE it! Haven't had much of a chance to tinker with it yet, but I can see it as a pretty decent way to "backdoor backup" your collections. Any time you can put them in more than one place, you are less likely to lose them in case of (gasp!) a disaster.
I wanted to make sure before I started writing about this new twist in our Kindle adventure, but it appears to look and act the same on either a Mac or Windows computer. If you decide to not import a collection (or collections) when you first install this new version, you can always go back and do it from the File menu. The software for Kindle for your computer is looking better every day. Even if you don't read a lot on your computer screen, it is a great way to keep track of what you have or haven't read yet.
Not such a happy story for the Kindle app for your iPad or iPhone... And the Kindle Cloud is still in its baby stages... no collections available for them yet. Maybe soon?

Ask Mrs. Wizard - Kindle Transferring
Transferring and Copying... Oh My!
Hi Cindy,
I spoke with you in January when I bought my Kindle, which I love! I bought
an iPad last month and I was able to get my Kindle library there without any
trouble, but alas not so with new laptop. I can’t even get my email contacts to
transfer, but that is another story!
Would you please let me know how I can transfer the Kindle file with as
little difficulty as possible?

About your Kindle books (library)... You don't actually "transfer" anything. Once you purchase a book on Amazon, it is in the Archives forever... You just need to install the Amazon software for whatever (iPad app, app for your pc, etc), then from that app, you can visit the archives in your Kindle library on Amazon and re download any books you want.
Now any books you got from places other than amazon are to be treated like files, and a lot there has to do with what kinds of files they are. If they are .mobi files, your Kindle software will open them ... so you can email your favorite books to yourself, then open that attachment on your iPad and it will ask if you want to open that file with your Kindle software app. Once you have opened the book, you can then delete the email. The book will be officially on your iPad.
Non Amazon book files can be opened and stored on your computer too... but you will need to either have the Kindle reader installed (in which case the only books you will be able to open will be .mobi files), or you can install the free mobipocket reader. That will give you quite a few more options for your non Amazon book files.
